目的探讨白介素-21(IL-21)对Jurkat细胞增殖的抑制作用及WASP家族Verprolin同源蛋白-1(WAVE-1)在急性白血病(AL)发病中可能的作用及意义。方法以不同浓度IL-21处理Jurkat细胞;通过MTT(噻唑兰比色法)检测观察IL-21对Jurkat细胞生长的影响;流式细胞仪观察24h后IL-21对细胞凋亡的影响;采用RT-PCR检测WAVE-1的表达。结果IL-21能明显抑制Jurkat细胞增殖,抑制作用呈剂量时间依赖效应(P<0.05);IL-21作用24h后,细胞凋亡随药物浓度增高而增加(P<0.05);WAVE-1 mRNA表达水平随IL-21处理浓度的增高而降低(P<0.05)。结论IL-21可以抑制人急性白血病细胞的生长和诱导其凋亡,其机制可能与降低WAVE-1的表达有关。
Objective To investigate the inhibitory effect of interleukin-21 (IL-21) on the proliferation of Jurkat cells and the possible role and significance of WASP family Verprolin homolog protein-1 (WAVE-1) in the pathogenesis of acute leukemia (AL). Methods Jurkat cells were treated with different concentrations of IL-21. The effect of IL-21 on Jurkat cells was observed by MTT assay. The effect of IL-21 on apoptosis was observed by flow cytometry. The expression of WAVE-1 was detected by RT-PCR. Results IL-21 could significantly inhibit the proliferation of Jurkat cells in a dose-time-dependent manner (P <0.05). After treated with IL-21 for 24 h, the apoptosis of Jurkat cells increased with the increase of drug concentration (P <0.05) The expression level decreased with the increase of IL-21 concentration (P <0.05). Conclusion IL-21 can inhibit the growth of human acute leukemia cells and induce their apoptosis. The mechanism may be related to the decrease of WAVE-1 expression.
