WRKY转录因子是植物界中存在的一类较大的基因家族,参与植物在生物和非生物逆境胁迫中的多种应答。为了研究在低温和高温等非生物胁迫对辣椒CaWRKY41基因表达的影响,以豫椒101为实验材料,在对辣椒CaWRKY41基因克隆的基础上,利用生物软件对其编码蛋白进行分析和构建进化树。结果表明,同源扩增获得的序列含有一个990 bp完整ORF框;对其编码蛋白分析发现,只含有一个WRKY结构域,属于WRKY41家族。亚细胞定位在细胞核中,分子进化树表明与茄属亲缘关系最近,相似性达到92%。荧光定量分析表明,与对照相比,低温、高温胁迫都能诱导CaWRKY41基因表达,表明CaWRKY41基因在应对低温和高温胁迫方面具有重要作用。
WRKY transcription factors are a family of larger genes present in the plant kingdom that are involved in multiple responses of plants to both abiotic and abiotic stresses. In order to study the effects of abiotic stresses such as low temperature and high temperature on CaWRKY41 gene expression in pepper, the pepper CaWRKY41 gene was cloned based on the cloning of pepper CaWRKY41 gene. The encoded protein was analyzed and the phylogenetic tree was constructed using biological software. The results showed that the sequence obtained by homologous amplification contained a complete ORF of 990 bp. The analysis of its encoded protein showed that it contained only one WRKY domain and belonged to the WRKY41 family. Subcellular localization in the nucleus, the molecular phylogenetic tree showed closest relationship with solanum, similar to 92%. Fluorescence quantitative analysis showed that CaWRKY41 gene expression was induced by both low temperature and high temperature stress compared with the control, indicating that CaWRKY41 gene plays an important role in coping with low temperature and high temperature stress.
