目的:观察雷公藤甲素(Triptolide,TP)对人正常肝细胞(L-O2)增殖的影响及机制研究。方法:采用CCK-8法、流式细胞仪检测细胞周期、细胞凋亡率的方法。结果:TP对L-O2细胞的增殖呈现剂量及时间依赖性抑制作用;TP对L-O2细胞周期的影响主要表现在将细胞阻断在S期;细胞的早期凋亡率与对照组相比有显著提高(P<0.01)。结论:TP对L-O2细胞具有增殖抑制作用,其机制可能与阻滞细胞周期和诱导L-O2细胞凋亡。
Objective: To observe the effects of triptolide (TP) on the proliferation of human normal liver cells (L-O2) and its mechanism. Methods: CCK-8 method was used to detect cell cycle and apoptosis rate by flow cytometry. Results: TP inhibited the proliferation of L-O2 cells in a time-and dose-dependent manner. The effect of TP on the cell cycle of L-O2 was mainly blocked in S phase. The early apoptosis rate of L-O2 cells compared with the control group There was a significant increase (P <0.01). CONCLUSION: TP can inhibit the proliferation of L-O2 cells in a dose-dependent manner, which may be related to arresting cell cycle and inducing L-O2 apoptosis.
