目的研究羊栖菜多糖(SFPS)对亲本果蝇生育能力的影响及其作用机制。方法采用85℃热水煮提、乙醇沉淀、氯化钙去除褐藻胶等多种工序提取SFPS;以黑腹果蝇为模式生物,分别以对照(0%)、低(0.033%)、中(0.167%)、高(0.500%)4个浓度持续喂食15 d,通过检测子代数量、体质量、每天羽化量评估不同浓度SFPS对果蝇生育能力的影响;荧光定量PCR(qRT-PCR)技术检测亲本果蝇生育和发育相关基因Vasa、Achi、Figla和子代果蝇mTOR的表达。结果与对照组比较,喂食SFPS能显著性地提高雌蝇和雄蝇的生育力,并且对其子代果蝇体质量、羽化率和雌雄比例也有后续的促进作用,低和中浓度SFPS分别对母本和父本果蝇的生育力促进效果最优;持续喂食SFPS后,亲本果蝇生殖发育相关基因在m RNA水平上显著提高;亲本喂食低、高浓度SFPS后,子代果蝇mTOR表达水平普遍升高,子代雌蝇变化更为显著。结论 SFPS通过调控果蝇生育相关基因的表达,提高亲本果蝇的生育能力,对子代果蝇的生长发育也有显著促进作用。
Objective To study the effect of SFPS on the fertility of the parent fruit fly and its mechanism. Methods SFPS was extracted by hot water boiling at 85 ℃, ethanol precipitation, and alginate removal by calcium chloride. SFDs were collected from control (0%), low (0.033%), 0.167%) and high (0.500%) for 15 days respectively. The effects of SFPS at different concentrations on the fertility of Drosophila melanogaster were evaluated by detecting the number of offspring, body weight and daily feathering amount. Quantitative PCR (qRT-PCR) The expression of the fertility and development-related genes Vasa, Achi, Figla and the progeny mTOR genes of the parents were examined. Results Compared with the control group, feeding SFPS significantly increased the fertility of female and male flies, and also had a subsequent promotion on the body weight, emergence rate and male-male ratio of their offsprings. SFPS of low and medium concentrations, respectively, The reproductive development-related genes of Drosophila melanogaster were significantly increased at m RNA levels after continuous feeding of SFPS. The mTOR expression levels of Drosophila Generally increased, the daughter females changes more pronounced. Conclusion SFPS can enhance the fertility of the fruit flies by controlling the expression of Drosophila related genes and also promote the growth and development of the fruit flies.
