目的 探讨髓过氧化物酶 (MPO)基因多态性与苯致血液毒性易感性的关联。方法 采用多聚酶链反应 -限制片段长度多态性 (PCR RFLP)技术和DNA测序法 ,分析 35例慢性苯中毒患者、46名苯作业工人和 2 6名健康工人的MPO基因转录起始位点上游 463位点等位基因分布情况 ,比较各组间的差别 ,探讨MPO基因多态性与苯中毒危险性的可能关联。结果 MPO基因上游 463位点存在G/G、G/A、A/A 3种基因型 ;G/G基因型在苯中毒病例组分布频率较苯接触组高 2 7.1 4 % ;MPO基因 463位点具G/G基因型个体发生苯中毒的危险性比G/A +A/A基因型增加 2 .8倍 (OR =2 .835 ,95 %CI:1 .0 65~ 7.549,P <0 .0 5) ;MPO基因多态性与苯中毒关联不存在性别差异。结论 在相同的苯作业暴露环境中 ,具G/G基因型个体较其他基因型发生苯中毒的风险高
Objective To investigate the association of myeloperoxidase (MPO) gene polymorphism with susceptibility to benzene-induced hematologic toxicity. Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing were used to analyze the transcriptional initiation sites of MPO in 35 patients with chronic benzene poisoning, 46 benzene workers and 26 healthy workers 463 loci distribution of alleles, the differences between the groups were compared to explore the possibility of MPO gene polymorphism and benzene poisoning risk associated. Results There were 3 G / G, G / A and A / A genotypes at 463 loci upstream of MPO gene. The distribution frequency of G / G genotype in benzene poisoning group was 21.44% higher than that in benzene exposure group and 463 loci The risk of benzene poisoning in individuals with G / G genotype was 2.8 times higher than that of G / A + A / A genotype (OR = 2.385, 95% CI: 1.65 ~ 7.549, P <0 .0 5). There was no gender difference in association between MPO gene polymorphism and benzene poisoning. Conclusions Individuals with G / G genotypes have a higher risk of benzene poisoning than other genotypes in the same benzene exposure environment
