血管生成素-2(angiopoietin-2,Ang2)是血管生成素家族的一员,在人类肿瘤发生和生长的血管生成中起了重要作用。Ang2在血管生成中的作用一般认为是Ang1激活的Tie2信号转导对血管成熟和血管稳定起关键作用而Ang2拮抗这种效应。Ang2与另一个重要血管生成因子VEGF-A以一种协调作用的方式调控血管生成。遗传学研究显示Ang2在淋巴管生成中也起重要作用。然而,新的证据表明,Ang2在人类肿瘤进展中的血管生成过程和肿瘤细胞侵袭性表型方面起更复杂的作用。本文综述了Ang2在血管生成过程中的一些最新研究进展,并讨论了针对肿瘤患者Ang2作为一个潜在抗血管生成治疗靶点和潜在抑制剂的应用。
Angiopoietin-2 (Ang2), a member of the angiogenin family, plays an important role in angiogenesis in the development and growth of human tumors. The role of Ang2 in angiogenesis It is generally thought that Ang1-activated Tie2 signaling plays a key role in vascular maturation and vascular stabilization, whereas Ang2 antagonizes this effect. Ang2 regulates angiogenesis in a coordinated manner with another important angiogenic factor, VEGF-A. Genetic studies have shown that Ang2 also plays an important role in lymphangiogenesis. However, new evidence suggests that Ang2 plays a more complex role in the angiogenic process and invasive phenotype of tumor cells in human tumor progression. This review summarizes some recent advances in Ang2 angiogenesis and discusses the use of Ang2 as a potential therapeutic and potential inhibitor of antiangiogenesis in cancer patients.
