目的:对中国11个不同产地的牛蒡进行ITS序列分析,为牛蒡DNA分子鉴定提供参考。方法:从11个不同产地的牛蒡中提取总DNA,用通用引物对ITS序列进行扩增,对扩增产物克隆测序。结果:牛蒡完整的ITS序列长度范围为641 bp,不同产地间无长度差异,GC含量为59.1%~59.6%之间,其中ITS1序列为256 bp,GC含量为58.6%~59.4%之间,其中ITS2序列为221 bp,GC含量为63.8%~64.7%之间。结论:11个不同产地的牛蒡ITS序列遗传距离为0.0000~0.0094,平均遗传距离为0.0024,相似度在99%以上,可以作为DNA分子鉴定的依据。
OBJECTIVE: To analyze the ITS sequences of Burdock in 11 different areas of China, and provide references for DNA molecular identification of burdock. Methods: Total DNA was extracted from burdock of 11 different producing areas. ITS sequences were amplified by universal primers, and the amplified products were cloned and sequenced. Results: The complete ITS sequence of Burdock was 641 bp in length with no difference in length between different origins. GC content ranged from 59.1% to 59.6%, including ITS1 sequence of 256 bp and GC content of 58.6% -59.4% ITS2 sequence was 221 bp with a GC content of 63.8% -64.7%. CONCLUSION: The ITS sequences of Burdock in 11 different habitats ranged from 0.0000 to 0.0094 with an average genetic distance of 0.0024 and a similarity of above 99%, which could be used as the basis for DNA molecular identification.
