为了更快捷、方便的检测朊病毒病,根据已有报道确定micro RNA-144-3p作为朊病毒病的生物标记,通过mi RBase数据库公布的micro RNA-142-3p的核酸序列,设计特异性标记有生物素的俘获探针和标记有FAM的检测探针,从而建立了纳米金偶联核酸试纸条的高效,快捷,可视化检测方法。结果表明:该检测方法具有良好的特异性和较高的灵敏度(0.005 nmol·L-1)。该检测试纸条的建立在朊病毒病预防,控制,诊断过程中具有一定实际意义。
In order to detect prion disease more quickly and conveniently, microRNA-144-3p has been reported as a biomarker of prion disease and a specific marker was designed based on the nucleic acid sequence of microRNA-142-3p published in mi RBase database Biotinylated capture probe and FAM-labeled detection probe, thus establishing an efficient, rapid and visual detection method of nano-gold conjugate nucleic acid strip. The results showed that this method has good specificity and high sensitivity (0.005 nmol·L-1). The establishment of the test strip in prion disease prevention, control, diagnosis has a certain practical significance.
