目的:检测了毒蕈样乙酰胆碱Ⅰ(MR1)和Ⅲ型受体(MR3)基因在大鼠胃粘膜的表达情况。方法:pSP73和pSP72质粒分别含有MR1和MR3cDNA,其用EcoRⅠ或HindⅢ线性化后,加入逆转录反应体系中制备地高辛标记的cRNA受体探针。胃粘膜MR1和MR3mRNA的定位检测采用原位杂交免疫组织化学方法。结果:MR1和MR3mRNA仅在大鼠胃粘膜固有层细胞表达,粘膜上皮细胞(腺体细胞)上无阳性的MR1和MR3mRNA杂交信号表达。结论:提示,胃粘膜上皮细胞(包括壁细胞和主细胞)可能缺乏乙酰胆碱受体,乙酰胆碱对胃酸分泌的刺激调节作用,可能是通过粘膜固有层的免疫细胞(淋巴细胞)介导的
OBJECTIVE: To detect the expression of muscarinic acetylcholine Ⅰ (MR1) and type Ⅲ receptor (MR3) genes in rat gastric mucosa. METHODS: Plasmids pSP73 and pSP72 contained MR1 and MR3 cDNA, respectively. After linearization with EcoRI or HindIII, they were added into the reverse transcription reaction system to prepare digoxigenin-labeled cRNA receptor probe. Gastric mucosa MR1 and MR3 mRNA localization detection using in situ hybridization immunohistochemistry. Results: MR1 and MR3 mRNA were only expressed in lamina propria of gastric mucosa in rats, and no positive MR1 and MR3 mRNA were expressed on mucosal epithelial cells (glandular cells). Conclusions: It is suggested that gastric epithelial cells (including parietal cells and major cells) may lack acetylcholine receptors, and acetylcholine may stimulate gastric acid secretion and may be mediated by immune cells (lymphocytes) in the lamina propria of mucosa
