[目的]观察热放疗对人肺腺癌耐药细胞株A549/DDP凋亡作用的影响。[方法]MTT法细胞毒性实验和克隆形成实验检测热放疗对A549/DDP细胞抑制作用和克隆形成的影响;TUNEL法检测热放疗对细胞凋亡作用的影响;透射电镜观察各组细胞形态学改变。[结果]细胞毒性实验和克隆形成实验显示热放疗对A549/DDP细胞生长有明显抑制作用(P<0.01);TUNEL法观察到热放疗有诱导A549/DDP细胞凋亡作用;电子显微镜观察到热放疗后A549/DDP出现细胞核固缩、变形,染色质浓聚、边集等凋亡现象。[结论]热放疗能诱导A549/DDP细胞凋亡。
[Objective] To observe the effect of thermal radiotherapy on the apoptosis of human lung adenocarcinoma cell line A549 / DDP. [Methods] MTT cytotoxicity assay and colony formation assay were used to detect the effect of heat-radiotherapy on A549 / DDP cell proliferation and colony formation. TUNEL assay was used to detect the effect of heat-radiotherapy on cell apoptosis. Transmission electron microscope was used to observe the morphological changes . [Results] Cytotoxicity experiments and colony formation experiments showed that heat radiotherapy significantly inhibited the growth of A549 / DDP cells (P <0.01). The apoptosis of A549 / DDP cells induced by heat radiotherapy was observed by TUNEL method. The results of electron microscopy showed that heat After radiotherapy A549 / DDP appeared nuclear pyknosis, deformation, chromatin condensation, edge set and other apoptosis. [Conclusion] Hyperthermia can induce the apoptosis of A549 / DDP cells.
