目的探讨JNK抑制剂CEP-1347对噪声损伤耳蜗的保护作用及意义。方法将27只Wistar大鼠随机分成3大组,A组:噪声组,B组:噪声+CEP-1347组,C组:噪声+盐水组,分别给予噪声暴露,B组在噪声暴露前给予皮下注射CEP-1347;C组同时间皮下注射等量生理盐水。在噪声暴露后的1、4及14d分别测试各组的ABR阈值,同时检测外毛细胞的凋亡情况。结果ABR阈值及外毛细胞凋亡数显示B组在1d时与A组比较差异无统计学意义(P>0.05),4d时听力较A组差异有统计学意义(P<0.05),用药后第14d听力与A组比较差异有统计学意义(P<0.01);C组在各时间点与A组比较差异无统计学意义(P>0.05)。结论应用JNK抑制剂CEP-1347进行干涉,可以减轻噪声造成的听力损失,JNK抑制剂的开发可能会成为N IHL防治的一个新的靶点和方向。
Objective To investigate the protective effects of JNK inhibitor CEP-1347 on cochlea in noise-damaged cochlea and its significance. Methods A total of 27 Wistar rats were randomly divided into three groups: group A: noise group, group B: noise + CEP-1347 group, group C: noise + saline group and noise exposure respectively. CEP-1347 was injected, while group C was injected subcutaneously with normal saline at the same time. The ABR threshold of each group was tested at 1, 4 and 14 days after noise exposure, and the apoptosis of outer hair cells was detected. Results The ABR threshold and the number of apoptotic cells showed that there was no significant difference between group B and group A on the 1st day (P> 0.05), and on the 4th day the difference was statistically significant (P <0.05) There was a significant difference between the 14th day of hearing and A group (P <0.01). There was no significant difference between A group and C group at each time point (P> 0.05). Conclusion Interference with JNK inhibitor CEP-1347 can reduce the noise-induced hearing loss, and the development of JNK inhibitors may be a new target and direction of NIRL prevention and treatment.
