目的探讨高糖环境对大鼠肾小球系膜细胞增殖、单核细胞趋化蛋白-1(MCP-1)的表达及NF-κB活化的影响。方法将体外培养大鼠肾小球系膜细胞(MC)分为4组:正常对照组(含5.6 mmol/L葡萄糖)、高糖1组(含15 mmol/L葡萄糖)、高糖2组(含20 mmol/L葡萄糖)和高糖3组(含25 mmol/L葡萄糖)。MTT法检测细胞的增殖情况,ELISA法检测MCP-1的表达,Western blotting法检测IκBα以反映NF-κB的活性。结果与正常对照组比较,高糖环境下24 h后细胞增殖明显,MCP-1表达明显增强(P<0.01)。高糖刺激30 min后,NF-κB结合蛋白IκBα明显降解,提示高糖可诱导MC NF-κB活化。结论高糖环境下大鼠肾小球系膜细胞的增殖及炎症因子MCP-1的高表达可能是通过NF-κB的活化来实现的。
Objective To investigate the effects of high glucose on the proliferation of mesangial cells, the expression of monocyte chemoattractant protein-1 (MCP-1) and the activation of NF-κB in rats. Methods Rat mesangial cells (MC) cultured in vitro were divided into 4 groups: normal control group (containing 5.6 mmol / L glucose), high glucose group 1 (containing 15 mmol / L glucose), high glucose group 2 Containing 20 mmol / L glucose) and high glucose 3 groups (containing 25 mmol / L glucose). Cell proliferation was detected by MTT assay, MCP-1 expression was detected by ELISA, and IκBα was detected by Western blotting to reflect the activity of NF-κB. Results Compared with the normal control group, the cell proliferation was obvious and the expression of MCP-1 was significantly increased after 24 h in high glucose environment (P <0.01). After 30 min of high glucose stimulation, the NF-κB binding protein IκBα was significantly degraded, suggesting that high glucose could induce the activation of MC NF-κB. Conclusion The proliferation of rat mesangial cells and the high expression of inflammatory factor MCP-1 in high glucose may be mediated through the activation of NF-κB.
