目的 :研究新的桂皮酰胺类衍生物 3 ,4 -二氯苯丙烯酰另丁胺 (AED880 1)对大鼠脑缺血 /再灌注损伤的影响。方法 :采用Pulsinelli四血管阻断大鼠脑缺血 /再灌注模型 ,观察了AED880 1对脑组织中水含量、氨基酸含量、6-keto-PGF1α和TXB2 含量 ,以及二者之比值的影响。结果 :一AED880 1能够降低脑缺血 /再灌注大鼠所引起的脑水含量增加。二采用氨基酸自动分析仪进行检测 ,发现脑缺血 /再灌注可致大鼠脑组织中谷氨酸 (Glu)含量增加 ,AED880 1可以使其降低 ,1 50mg/kg作用非常显著 (P <0 .0 1 )。对所测其它氨基酸含量虽有所影响 ,但均无统计学意义。三AED880 1可增加脑缺血 /再灌注大鼠脑中 6-keto -PGF1α含量 ,减少TXB2 含量 ;因而增加二者之比值。结论 :AED880 1对脑缺血 /再灌注具有保护作用 ,这种作用可能与其降低脑中Glu含量 ,增加脑中PGI2 含量和减少TXA2 含量 ,并提高二者之比值有关
Objective: To study the effect of AED880 1, a new cinnamate derivative, on cerebral ischemia / reperfusion injury in rats. Methods: Pulsinelli four-vessel occlusion model was used to observe the effects of AED880 1 on water content, amino acid content, 6-keto-PGF1α and TXB2 content in brain tissue and the ratio between the two. Results: One AED880 1 can reduce the increase of brain water content induced by cerebral ischemia / reperfusion in rats. Second, the amino acid analyzer was used to detect. Cerebral ischemia / reperfusion induced the increase of glutamate (Glu) content in brain tissue of rats. AED880 1 could reduce it, and the effect of 150 mg / kg was very significant (P <0. 0 1). The measured other amino acid content although the impact, but no statistically significant. Three AED880 1 can increase the content of 6-keto-PGF1α in the brain of rats with cerebral ischemia / reperfusion and reduce the content of TXB2; thus increasing the ratio of the two. CONCLUSION: AED880 1 has a protective effect on cerebral ischemia / reperfusion. This effect may be related to its effect on reducing the level of Glu in the brain, increasing PGI2 in the brain, decreasing TXA2 content, and increasing the ratio between the two
