目的研究灯盏乙素对脂多糖(lipopolysaccharide,LPS)诱导BV2细胞株炎症细胞因子表达的影响,探讨灯盏乙素抗炎作用机制。方法通过荧光素酶报告基因检测核转录因子NF-κB活性,并利用LPS刺激使BV2细胞和通过RT-PCR和蛋白免疫法检测TNF-α和IL-1β的mRNA及蛋白含量,研究灯盏乙素的抗炎作用。实验分为正常组、LPS刺激(1μg/ml)的模型组、灯盏乙素组(0.1、1、10和100μg/ml)。结果灯盏乙素预处理(1~100μg/ml)可抑制LPS引起pNF-κB 293的核转录因子NF-κB激活(P<0.05或P<0.01);灯盏乙素预处理(1~100μg/ml)能够抑制LPS引起BV2的TNF-α和IL-1βmRNA和蛋白含量升高(P<0.01)。结论灯盏乙素具有抗炎作用,其抗炎作用与抑制核转录因子NF-κB、抑制炎症细胞因子表达有关。
Objective To investigate the effect of scutellarin on the expression of inflammatory cytokines in BV2 cells induced by lipopolysaccharide (LPS) and explore the anti-inflammatory mechanism of scutellarin. Methods The NF-κB activity of NF-κB was detected by luciferase reporter assay. The mRNA and protein levels of TNF-α and IL-1β in BV2 cells were detected by LPS stimulation and the mRNA and protein levels of TNF-α and IL-1β were detected by RT- Anti-inflammatory effect. The experiment was divided into normal group, LPS stimulation (1μg / ml) model group, scutellarin group (0.1, 1, 10 and 100μg / ml). Results Scutellarin preconditioning (1 ~ 100μg / ml) could inhibit the activation of NF-κB induced by LPS (P <0.05 or P <0.01) ) Could inhibit the increase of BV2 TNF-α and IL-1βmRNA and protein levels in LPS (P <0.01). Conclusion Scutellarin has anti-inflammatory effects, and its anti-inflammatory effects are related to inhibition of nuclear factor-κB and inhibition of inflammatory cytokines expression.
