背景:高脂血症的长期存在可促进动脉粥样硬化(atherosclerosis,AS)的形成和发展,在此过程中生长因子所起的作用越来越受重视,但对血管内皮生长因子(vascular endothelial growth factor, VEGF)的作用还不十分清楚。 目的:探讨VEGF在AS形成过程中的作用。 设计:随机对照的实验研究。 地点、材料和干预:本实验在沈阳体育学院研究生实验室完成。实验动物为C57BL/6J(易感AS型)纯系小黑鼠20只。实验动物被随机分两组:对照组10只,脂肪乳组10只。对照组喂以常备标准饲料;脂肪乳组除正常喂养外,每日以脂肪乳灌喂方式按10mL/kg进行高脂膳食。 主要观察指标:高脂血症与非高脂血症小鼠主动脉组织光镜下形态学改变、主动脉壁的VEGF蛋白表达、血脂水平比较。 结果:对照组主动脉壁VEGF蛋白表达均为阴性,仅外膜残存脂质VEGF阳性。脂肪乳组血栓中、局部增生的新内膜均有VEGF蛋白表达。血脂测定结果:脂肪乳组的总胆固醇、三酰甘油、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,HDL-C)分别为(4.97±0.10),(2.24±0.26),(2.93±0.17)mmol/L较对照组[分别为(4.17±0.29),(1.22±0.10),(2.44±0.33)mmol/L]明显升高(t=2.364,2.335,2.939,P<0.01);脂肪乳组HDL-C[(1.29±0.24)mmol/L]较对照组[(1.50±0.16)mmol/L]
BACKGROUND: The long-term existence of hyperlipidemia promotes the formation and development of atherosclerosis (AS). In this process, the role of growth factor is more and more emphasized. However, the effect of vascular endothelial growth factor growth factor, VEGF) role is not yet clear. Objective: To investigate the role of VEGF in AS formation. Design: Randomized controlled experimental study. Location, Materials and Interventions: This lab was completed at the Graduate School of Shenyang Institute of Physical Education. Experimental animals C57BL / 6J (susceptible AS type) 20 black mice. Experimental animals were randomly divided into two groups: the control group of 10, 10 fat emulsion group. The control group fed normal standard feed; fat milk group in addition to normal feeding, fat milk daily feeding method by 10mL / kg for high-fat diet. MAIN OUTCOME MEASURES: Morphological changes of aorta in hyperlipidemic and non-hyperlipidemic mice, expression of VEGF protein in aortic wall, and blood lipid levels were compared. Results: The expression of VEGF protein in the aorta wall of the control group was negative, and only the peripheral adventitial lipid VEGF was positive. Among the thrombus in the fat emulsion group, VEGF expression was found in the neointimal hyperplasia. The results of lipid profile: Total cholesterol, triglyceride and low density lipoprotein cholesterol (HDL-C) were 4.97 ± 0.10, 2.24 ± 0.26, 2.93 ± 0.17, / L compared with the control group [(4.17 ± 0.29), (1.22 ± 0.10) and (2.44 ± 0.33) mmol / L, respectively C [(1.29 ± 0.24) mmol / L] compared with control group [(1.50 ± 0.16) mmol / L]
