目的探讨乌司他丁对急性肺损伤(ALI)大鼠肺组织中Toll样受体4(TLR4)及肺泡灌洗液中的髓样细胞触发受体1(TREM-1)和TREM-2表达的影响。方法将72只SD大鼠随机均分为三组。LPS组、LU组大鼠腹腔注射脂多糖(LPS)5mg/kg建立ALI模型;C组腹腔注射等容量生理盐水。LU组同时腹腔注射乌司他丁10万U/kg。每组给药后4、8、12和24h分别处死6只大鼠,采用RT-PCR半定量检测肺组织TLR4mRNA表达,ELISA法检测肺泡灌洗液中TREM-1和TREM-2表达。结果 LPS组各时间点的TLR4 mRNA、TREM-1和TREM-2表达均高于C组(P<0.05);LU组TLR4mRNA和TREM-1表达均低于LPS组(P<0.05),而TREM-2表达高于LPS组(P<0.05)。结论乌司他丁对LPS诱导的ALI有保护作用,可能与下调TLR4 mRNA、TREM-1表达和上调TREM-2表达有关。
Objective To investigate the effects of ulinastatin on the expression of TRAIL-1 and TREM-2 in lung tissue of acute lung injury (ALI) rats and TLR4 in bronchoalveolar lavage fluid Impact. Methods 72 SD rats were randomly divided into three groups. Rats in LPS group and LU group were given intraperitoneal injection of lipopolysaccharide (LPS) 5mg / kg to establish ALI model. Rats in group C were injected intraperitoneally with normal saline. LU group intraperitoneal injection of ulinastatin 100,000 U / kg. Six rats were sacrificed at 4, 8, 12 and 24 h after administration. The expression of TLR4 mRNA in lung tissue was detected by semi-quantitative RT-PCR and the expression of TREM-1 and TREM-2 in bronchoalveolar lavage fluid by ELISA. Results The expressions of TLR4 mRNA, TREM-1 and TREM-2 at each time point in LPS group were significantly higher than those in C group (P <0.05). The expression of TLR4 mRNA and TREM-1 in LPS group was lower than that of LPS group (P <0.05) -2 was higher than that in LPS group (P <0.05). Conclusion Ulinastatin has a protective effect on LPS-induced ALI, which may be related to the down-regulation of TLR4 mRNA, TREM-1 expression and up-regulation of TREM-2 expression.
