目的研究大鼠在脓毒症时组织因子(TF)的表达,探讨盐酸戊乙奎醚(penehyclidine hydr ochl ordie,PHC)是否通过核因子-κB(NF-κB)途径抑制组织因子的增加。方法将30只大鼠随机分为假手术组(A组)、手术组(B组)、PHC组(C组)、四氢化吡咯二硫代氨基甲酸酯(PTDC)组(D组)、PHC+PTDC组(E组)。B、C、D、E组用盲肠结扎穿刺法(CLP)造模,A组除结扎穿刺外处理同其他组。造模后12h处死大鼠,留取右肺上叶组织约0.5cm3测TF,右肺中叶组织约0.5cm3测NF-κB。结果12h时A、B、C、D、E组,TF及NF-κB比较,B组较A组NF-κB升高,TF增加;C、D组较B组NF-κB水平下降,TF减少;较A组NF-κB升高,TF增加。E组较A组NF-κB、TF水平没有变化。结论 TF水平的升高与NF-κB含量的增加有一定相关性,TF可能至少有部分通过NF-κB途径介导合成,PHC可以有效抑制NF-κB活化,TF表达。
Objective To investigate the expression of tissue factor (TF) during sepsis in rats and to explore whether penehyclidine hydrocholine (PHC) inhibits the increase of tissue factor through the nuclear factor-κB (NF-κB) pathway. Methods Thirty rats were randomly divided into sham operation group (A group), operation group (B group), PHC group (C group), PTDC group (D group) PHC + PTDC group (group E). Groups B, C, D and E were established by cecal ligation and puncture (CLP). Group A was treated with the same method except for ligation and puncture. Rats were sacrificed at 12h after modeling. Tissues from the upper lobe of right lung were collected and measured for about 0.5cm3. NF-κB was measured in the right middle lobe of about 0.5cm3. Results Compared with group A, B, C, D, E, TF and NF-κB, the level of NF-κB in group B increased and TF increased at 12h; the level of NF-κB and the decrease of TF in group C and group D Compared with group A, NF-κB increased and TF increased. E group compared with A group of NF-κB, TF levels did not change. Conclusions The increase of TF level has a certain correlation with the increase of NF-κB. TF may be at least partially mediated by NF-κB pathway. PHC can inhibit the activation of NF-κB and the expression of TF.
