目的观察分泌表达NT4-TAT-6×His-VHLβ肾癌抑制融合肽的重组腺伴随病毒作用于肾癌鸡胚移植瘤的疗效。方法将人肾癌细胞系RLC-310接种到鸡胚绒毛尿囊膜上。选取成瘤较好鸡胚,分别用rAAV/NT4-TAT-6×His-VHLβ(A组)、空病毒(B组)、PBS缓冲液(C组)进行干预,观察移植瘤生长及各组移植瘤的大小,采用HE染色及免疫荧光法比较各组的作用效果。结果 A组移植瘤生长速度明显慢于其他两组;A组移植瘤体积与B、C两组比较差异有统计学意义(P<0.05);HE染色结果显示,A组鸡胚移植瘤生长受到抑制;荧光显微镜可观察到A组荧光明亮(++),对照组未观察到荧光(-)。结论 rAAV/NT4-TAT-6×His-VHLβ对肾癌鸡胚移植瘤有抗肿瘤效应,为肾癌的基因治疗研究提供了一种新的途径。
Objective To observe the therapeutic effect of recombinant adeno-associated virus secreting and expressing NT4-TAT-6 × His-VHLβ renal carcinoma on fusion of chick embryo xenografts in renal cell carcinoma. Methods Human renal carcinoma cell line RLC-310 was inoculated into chick chorioallantoic membrane. The tumorigenic embryos were selected and the intervention of rAAV / NT4-TAT-6 × His-VHLβ (group A), empty virus (group B) and PBS buffer (group C) The size of the xenografts, HE staining and immunofluorescence were used to compare the effect of each group. Results The growth rate of transplanted tumor in group A was significantly slower than that in the other two groups. The volume of transplanted tumor in group A was significantly lower than that in group B and C (P <0.05). The HE staining showed that the growth of transplanted tumor in group A was affected Inhibition; Fluorescence microscope can be observed in group A fluorescence bright (++), the control group was not observed fluorescence (-). Conclusion The rAAV / NT4-TAT-6 × His-VHLβ has antitumor effect on the chick embryo xenografts in renal cell carcinoma, which provides a new approach for the gene therapy of renal cell carcinoma.
