目的:建立血清中硫酸奈替米星的高效液相色谱间接光度检测法(HPLCIDP)。方法:以烟酰胺为检测剂,庚烷磺酸钠为反离子,固定相SpherisorbC18,流动相为甲醇pH2的磷酸缓冲液(20∶80),内含烟酰胺0.4mmol·L-1,庚烷磺酸钠5mol·L-1。测定了4例患者单剂量im后的血清浓度并计算药动学参数。结果:线性范围0.5mg·L-1~15.0mg·L-1,线性回归方程c=0.855-4.384×10-6H,r=0.998。回收率平均为93.27%,RSD日内平均为4.95%,日间平均为5.71%。结论:本法操作简便,灵敏度高,适于作为血清中硫酸奈替米星的检测方法
Objective: To establish a method for the determination of netilmicin sulfate in serum by HPLC-IDP. Methods: Nicotinamide was used as the detection reagent and sodium heptane sulfonate as the counter ion. The stationary phase was Spherisorb C18. The mobile phase consisted of methanol-pH2 phosphate buffer (20:80) containing nicotinamide 0.4 mmol·L-1, Heptane sulfonate 5mol · L-1. Four patients were measured after a single dose im serum concentration and calculated pharmacokinetic parameters. Results: The linear range was 0.5 mg · L-1 ~ 15.0 mg · L-1. The linear regression equation was 0.855-4.384 × 10-6H, r = 0.998. The average recovery was 93.27%. The RSD was 4.95% on average and the average daytime was 5.71%. Conclusion: The method is simple, sensitive and suitable for the detection of netilmicin sulfate in serum
