目的 用重组质粒pc D N A3 - P30 免疫 B A L Bc 小鼠, 观察其 D N A 免疫所诱导的小鼠体液免疫反应和保护性作用。方法 大量制备质粒 D N A, 肌肉注射免疫小鼠。 E L I S A 测定 Ig G 抗体滴度; 免疫鼠血液组织 P30 基因 P C R 扩增; 弓形虫攻击感染。结果 用 E L I A S 法检测的抗体 Ig G 滴度1∶2560 , 免疫后3 周及6 个月从免疫鼠血液组织中检测到 P30 基因;攻击感染实验组的存活时间较对照时间延长( P< 005) , 结论 重组质粒pc D N A3 - P30 免 B A L Bc 小鼠可诱导一定的体液免疫应答, 对抗攻击感染具有一定的保护性。
OBJECTIVE: To investigate the humoral immune response and protective effect of mice immunized with D N A against B A L Bc mice by recombinant plasmid pc D N A3 - P30. Methods Plasmid D N A was prepared in large quantities and immunized intramuscularly. E L I S A Ig G antibody titers were measured; P C R amplification of P30 gene in the blood of immunized mice; Toxoplasma gondii infection. Results The IgM titer 1:2560 was detected by ELIAS method. The P30 gene was detected from the blood of immunized mice at 3 weeks and 6 months after immunization. The survival time of experimental group was longer than that of control group P <005). Conclusions The recombinant plasmid pcDNA3 - P30 - free B L Bc mice can induce a certain humoral immune response and have some protection against attack of infection.
