目的研究转HBsAg基因可食用疫苗苜蓿(Edible Vaccine Alfalfa Transformated with HBsAg Gene,EVATHG)对免疫功能的影响。方法将EVATHG按0.5 g/kg~2.0 g/kg给小鼠灌胃30 d后,测定各项免疫学指标并进行病理组织学检查。结果与阴性对照组比较,EVATHG可明显提高小鼠脾淋巴细胞转化能力、增加小鼠血清溶血素抗体积数值,差异均有统计学意义(P<0.05)。而变态反应性、溶血空斑数、腹腔巨噬细胞吞噬鸡红细胞能力、NK细胞杀伤活性、碳廓清能力和体重增加则变化不明显,各组间差异无统计学意义(P>0.05)。与野生型苜蓿组相比,各项免疫学指标差异均无统计学意义(P>0.05)。病理组织学检查未见EVATHG对组织器官的损伤作用。结论与野生型苜蓿相同,EVATHG亦能促进T淋巴细胞的增殖,增加血清溶血素抗体水平,对小鼠的免疫功能无抑制作用。在本实验条件下,未见EVATHG有免疫毒性。
Objective To investigate the effect of Edible Vaccine Alfalfa Transformated with HBsAg Gene (EVATHG) on immune function. Methods EVATHG was intragastrically administered to mice at a dose of 0.5 g / kg ~ 2.0 g / kg for 30 days. The immunological indexes were measured and pathological examination was performed. Results Compared with the negative control group, EVATHG could significantly increase the ability of splenic lymphocyte transformation in mice and increase the antibody titer of serum hemolysin in mice (P <0.05). Alteration, hemolytic plaque number, peritoneal macrophage phagocytosis of chicken erythrocytes, NK cell killing activity, carbon clearance ability and weight gain did not change significantly, with no significant difference between the groups (P> 0.05). Compared with the wild-type alfalfa group, the immunological indexes had no significant difference (P> 0.05). No histopathological examination of EVATHG damage to tissues and organs. Conclusion As with wild-type alfalfa, EVATHG can also promote the proliferation of T lymphocytes and increase the level of serum hemolysin antibody, which has no inhibitory effect on immune function in mice. Under the experimental conditions, no EVATHG immunotoxicity.
