目的 探讨血小板第 4因子 (PF4)对小鼠 7 5Gyγ射线全身照射所致急性放射损伤的防护作用及机理。方法 小鼠照射前 2 0h于腹腔内注射PF4两次 ,间隔 6h ,每次剂量 2 0~ 6 0μg/kg。观察动物的 30d存活情况及几种造血指标。 结果 30d存活率对照组和防护组分别为 0 %和 5 0 %~ 6 0 % (P <0 0 1) ;防护组的CFU GM集落形成数、骨髓细胞DNA含量、骨髓有核细胞数、脾结节形成数、脾体比、胸体比等均较照射对照组增加 ,经统计学处理差异有显著性 (P <0 0 5 )或有非常显著性 (P <0 0 1)。结论 PF4对小鼠急性放射损伤有防护作用 ,其机理可能同它保护和促进造血功能恢复有关。
Objective To investigate the protective effect and mechanism of platelet factor 4 (PF4) on acute radiation injury induced by whole-body radiation of 75 Gy in mice. Methods Mice were injected intraperitoneally twice with PF4 2 hours before irradiation, with a dose of 20 ~ 60μg / kg every 6 hours. The animals were observed 30d survival and several hematopoietic indicators. Results The survival rate of control group and protective group at 30d were 0% and 50% ~ 60%, respectively (P <0.01). The CFU GM colony formation, the DNA content of bone marrow cells, the number of bone marrow nucleated cells, The number of nodules, the ratio of spleen to body and the ratio of thoracic body increased compared with that of the irradiated group. The difference was statistically significant (P <0.05) or significant (P <0.01). Conclusion PF4 has a protective effect on acute radiation injury in mice and its mechanism may be related to its protection and promotion of hematopoietic function recovery.
