目的 观察雷公藤甲素 (T10 )对平滑肌细胞 (SMCs)细胞周期进程的影响和诱导凋亡作用。方法 体外主动脉SMCs培养和流式细胞仪技术。结果 T10 与指数增殖期SMCs共育后 ,可引起核染色质浓缩、聚集 ,细胞膜内陷 ,凋亡小体形成等凋亡形态学改变。流式细胞仪分析 ,在G1峰前出现凋亡峰 ,峰值随T10 浓度增加而升高 ;G1期细胞进入S期并阻滞于S期 ;G1、M期细胞比例下降而S期升高 ;凋亡分子Fas表达明显升高。结论 T10 可以诱导指数增殖期SMCs凋亡
Objective To observe the effect of triptolide (T10) on the cell cycle progression and the induction of apoptosis of smooth muscle cells (SMCs). Methods In Vitro Aortic SMC Culture and Flow Cytometry. Results T10 and exponential proliferation of SMCs co-education, can cause nuclear chromatin condensation, aggregation, cell membrane invaginations, apoptotic bodies such as apoptotic morphological changes. Flow cytometry analysis showed that the peak of apoptosis occurred before G1 peak and the peak value increased with the increase of T10 concentration. The cells in G1 phase entered S phase and arrested in S phase. The proportion of cells in G1 and M phase decreased but increased in S phase. Apoptotic molecules Fas expression was significantly increased. Conclusion T10 can induce apoptosis of exponential proliferation of SMCs
