目的探讨胎盘组织中ADAM10及其底物E-cad、受体Flt-1的表达与子痫前期发病的关系。方法以厦门大学附属第一医院2011-2015年住院分娩的45例重度子痫前期病患为研究对象设为子痫前期组,并以同期50例正常妊娠孕妇为对照设为正常妊娠组,分别检测两组胎盘组织中3种因子mRNA及蛋白的表达。结果子痫前期组ADAM10和Flt-1mRNA表达均高于正常妊娠组,其蛋白质水平也均高于正常妊娠组,而E-cadmRNA和蛋白质均低于正常妊娠组,差异均有统计学意义(P<0.01)。结论重度子痫前期胎盘组织中ADAM10、Flt-1过度表达及E-cad低表达可能与子痫前期的发生和发展密切相关。
Objective To investigate the relationship between the expression of ADAM10, E-cad and its receptor Flt-1 in placenta and the pathogenesis of preeclampsia. Methods Forty-five patients with severe preeclampsia who were hospitalized and delivered from 2011 to 2015 in the First Affiliated Hospital of Xiamen University were enrolled as the preeclampsia group and 50 normal pregnant women as the control group were set as normal pregnancy group The mRNA and protein expression of three factors in placenta were detected. Results The expression of ADAM10 and Flt-1 mRNA in preeclampsia group were higher than those in normal pregnancy group, and their protein levels were also higher than those in normal pregnancy group, while the expression of E-cadherin and protein were lower than those in normal pregnancy group (P <0.01). Conclusion The over-expression of ADAM10 and Flt-1 and the low expression of E-cad in placenta of severe preeclampsia may be closely related to the occurrence and development of preeclampsia.
