目的:为优良菘蓝四倍体株系选育提供参考。方法:HPLC法分析靛蓝和靛玉红的含量,采用苯酚-硫酸法测定多糖含量,利用氨基酸分析仪测定氨基酸含量。结果:菘蓝四倍体株系根中靛蓝、靛玉红含量大多数高于二倍体株系,而叶中靛蓝和靛玉红含量不高于二倍体;四倍体株系根中的多糖含量均低于二倍体,其中以四倍体株系DB1、DB10和DB15根中多糖含量较高,DB6、DB13和DB15叶中多糖含量最高;检测出17种氨基酸,其中必需氨基酸6种,而精氨酸和脯氨酸含量为最高。其中DB7、DB8根和叶中的精氨酸含量均高;DB1、DB7和DB12脯氨酸含量高;DB7、DB9和DB12叶中总氨基酸含量最高。结论:菘蓝不同四倍体株系有效成分表现不完全一致,生产上根据实际应用情况进行选用。
Objective: To provide references for the breeding of tetraploid plants with excellent blue. Methods: The contents of indigo and indirubin were analyzed by HPLC. The content of polysaccharides was determined by phenol-sulfuric acid method. The content of amino acids was determined by amino acid analyzer. Results: The contents of indigo and indirubin in roots of tetraploid plants were higher than those in diploid plants, while the content of indigo and indirubin in leaves was not higher than that of diploid plants. Of polysaccharides were lower than that of diploid. Among them, the content of polysaccharides in DB1, DB10 and DB15 of tetraploid strains was the highest, and the content of polysaccharides in DB6, DB13 and DB15 was the highest. Seventeen kinds of amino acids were detected, of which essential amino acids 6 Species, arginine and proline content is the highest. Among them, the contents of arginine in roots and leaves of DB7, DB8 were all high; the contents of proline in DB1, DB7 and DB12 were high; the contents of total amino acids in DB7, DB9 and DB12 were the highest. CONCLUSION: The active constituents of different tetraploid strains of Isatis indigotica are not exactly the same, and their production is selected according to the actual application.
