目的:对甘草酸二铵结肠定位微囊(DCSM)的处方和工艺进行研究。方法:流化床包衣法制备甘草酸二铵结肠定位微囊,以释放度为指标,确定果胶与乙基纤维素水分散体的比例及包衣增重,以微囊收率为指标,采用U8(84)均匀设计试验优化工艺参数。结果:确定果胶与乙基纤维素水分散体的比例为1∶4,包衣增重43%;优化包衣工艺为包衣液浓度4%,进风温度55℃,喷雾压力2.6 kg,输液速度0.6 mL·min-1。结论:DCSM具有良好的体外结肠定位释药特性,可进一步进行体内释药行为考察。
Objective: To study the prescription and technology of diammonium glycyrrhizinate colon-localized microcapsules (DCSM). Methods: Ammonium dihydrogen glycyrrhizinate colon microcapsules were prepared by fluidized bed coating method. The release ratio was used as an index to determine the ratio of pectin to ethyl cellulose aqueous dispersion and the weight gain of coating. , Using U8 (84) uniform design test to optimize process parameters. Results: The ratio of pectin and ethyl cellulose aqueous dispersion was 1: 4 and the weight gain of coating was 43%. The optimal coating process was 4% concentration of coating solution, air inlet temperature 55 ℃, spray pressure 2.6 kg, Infusion speed 0.6 mL · min-1. CONCLUSION: DCSM has good colon-specific release characteristics in vitro and can be further investigated in vivo.
