用秋水仙碱处理二倍体甜菜不能直接得到纯四倍体,只能得到嵌合体,采用常规技术从嵌合体获得足够量的纯四倍体种子至少需4年时间.应用组织培养技术将嵌合体上的四倍体器官做外植体,接种到培养基上培养成苗并达到增殖生根,当年收获了相当数量的母根.将母根经3~5℃化处理,第2年便可获得纯四倍体甜菜种子。
Treatment of diploid beets with colchicine did not result in pure tetraploids, but only chimeras, obtaining a sufficient amount of pure tetraploid seeds from the chimera for at least 4 years using conventional techniques. Tissue culture technique was used to make tetraploid organs on the chimera as explants. The explants were inoculated into the culture medium to cultivate the seedlings and proliferate the roots. In the same year, a considerable number of maternal roots were harvested. The mother roots by 3 ~ 5 ℃ treatment, the second year can be pure tetraploid beet seeds.
