目的:探讨肿瘤坏死因子-α(tumor necrosis factor,TNF-α)、白细胞介素-6(interleukin-6,IL-6)在糖尿病大鼠骨骼肌中的表达及其与细胞凋亡的关系。方法:将Wistar大鼠分为实验组与对照组各8只,实验组给予高脂、高糖饮食8周后腹腔内注射小剂量链脲佐菌素(30mg/kg),对照组喂养标准大鼠饲料。第13周末处死大鼠,取各组大鼠股四头肌检测TNF-α及IL-6蛋白表达及bcl-2和Caspase-3的表达,并分析与以上二者的相关性。结果:与对照组比较,实验组大鼠骨骼肌中TNF-α与IL-6的表达水平均升高(P<0.01),bcl-2表达明显低于对照组(P<0.01),TNF-α、IL-6与bcl-2的表达呈负相关性(r=-0.876,r=-0.823,P<0.05);Caspase-3表达明显高于对照组(P<0.01),TNF-α、IL-6与Caspase-3的表达呈正相关性(r=0.713,r=0.692,P<0.05)。结论:糖尿病骨骼肌病变时炎症与凋亡同时发生,并可能共同导致胰岛素抵抗。
Objective: To investigate the expression of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in skeletal muscle of diabetic rats and its relationship with apoptosis. Methods: Wistar rats were divided into experimental group and control group (n = 8). Experimental group were given high-fat and high-sugar diet for 8 weeks and then intraperitoneal injection of low-dose streptozotocin (30mg / kg) Rat feed. At the end of the thirteenth week, the rats were sacrificed and the expressions of TNF-α and IL-6 protein, bcl-2 and Caspase-3 were detected by quadriceps femoris in each group. The correlation between them was analyzed. Results: Compared with the control group, the expression of TNF-α and IL-6 in skeletal muscle of experimental group were significantly increased (P <0.01), the expression of bcl-2 was significantly lower than that of control group α, IL-6 and bcl-2 were negative correlated (r = -0.876, r = -0.823, P <0.05); the expression of Caspase-3 was significantly higher than that of the control group There was a positive correlation between the expression of IL-6 and Caspase-3 (r = 0.713, r = 0.692, P <0.05). CONCLUSION: Inflammation and apoptosis occur simultaneously in diabetic skeletal muscle and may lead to insulin resistance.
