目的探讨不同推荐剂量参芎化瘀胶囊预处理对全脑缺血再灌注大鼠海马VEGF和GAP-43的影响。方法将125只成年SD雄性大鼠随机分为假手术组(n=25)、模型组(n=25)、参芎化瘀胶囊低、中、高剂量组(240,360,480mg/kg)。制备全脑缺血再灌注动物模型,取24,48,72,168,336 h 5个时间点。HE染色观察海马CA1区组织学改变,免疫组化法检测VEGF、GAP-43的表达。平衡木法测定行为学评分。结果与假手术组比较,模型组神经元形态明显受损,细胞密度减少明显,行为学评分升高,VEGF、GAP-43表达升高(P<0.05);和模型组比较,参芎化瘀胶囊干预各组神经元细胞数量增加,行为学评分降低,VEGF先升高后降低,GAP-43升高明显(P<0.05),以高剂量组最为显著(P<0.05)。结论参芎化瘀胶囊预处理对全脑缺血再灌注大鼠神经元损伤有保护作用,其中高剂量最为显著,其机制可能和促进VEGF、GAP-43的表达有关。
Objective To investigate the effects of different doses of Shenxionghuayu capsule on VEGF and GAP-43 in hippocampus of global cerebral ischemia-reperfusion rats. Methods 125 adult SD male rats were randomly divided into sham operation group (n = 25), model group (n = 25) and Shenxionghuayu capsule low, medium and high dose groups (240,360 and 480 mg / kg). The whole brain ischemia-reperfusion animal model was established, taking 24,48,72,168,336 h 5 time points. The histological changes of hippocampal CA1 area were observed by HE staining. The expression of VEGF and GAP-43 were detected by immunohistochemistry. Behavioral score of balance beam method. Results Compared with the sham-operated group, the morphological changes of neurons in the model group were significantly impaired, the cell density decreased significantly, and the behavioral scores increased, while the expression of VEGF and GAP-43 increased (P <0.05). Compared with the model group, The number of neurons in each group increased, the behavior score decreased, VEGF increased first and then decreased, GAP-43 increased significantly (P <0.05), and the highest dose group was the most significant (P <0.05). Conclusion Shenxionghuayu capsule preconditioning can protect neurons from injury after global cerebral ischemia and reperfusion in rats. The high dose is the most significant. The mechanism may be related to the promotion of the expression of VEGF and GAP-43.
