树突状细胞(dendritic cells,DC)是一种抗原递呈细胞,其最显著的特点是能有效刺激静息的T淋巴细胞,诱发初次免疫应答.DC来源于骨髓细胞,在正常组织中含量很少,在体外可用GM-CSF联合IL-4能从CD14~+单个核细胞培养出DC,细胞因子对DC的分化成熟和抗原递呈功能具有重要的调节作用.我们在体外成功的建立了16例肿瘤患者的自体DC细胞.血标本的来源为中晚期消化道肿瘤患者,年龄25~60岁,平均年龄52岁,其中男性9例,女性7例.实验方法:肿瘤患者外周血经CS-3000血细胞分离机采集,经Ficol淋巴细胞分层液密度梯度离心分离获得单个核细抱,计数
Dendritic cells (DCs) are antigen-presenting cells. Their most prominent feature is their ability to stimulate resting T lymphocytes and induce a primary immune response. DCs are derived from bone marrow cells and are present in normal tissue. Rarely, in vitro, GM-CSF and IL-4 can be used to culture DCs from CD14~+ mononuclear cells. Cytokines play an important role in the regulation of DC maturation and antigen-presenting function. We have successfully established in vitro. 16 autologous DC cells from tumor patients.
