目的 观察新型基因工程人肿瘤坏死因子α(nrhT NF α) ,对小鼠移植性肿瘤 (肉瘤S180、肝癌H2 2、黑素瘤B16和Lewis肺癌 )生长的抑制作用。方法以 3种剂量 (37 5、75和 15 0万U/kg体重 )的nrhTNF α肌肉注射小鼠 ,每天 1次 ,连续 8d。通过称量所形成的肿瘤重量 ,观察nrhTNF α对瘤细胞株的抑制作用。结果肿瘤抑制率计算结果显示 ,nrhTNF α对 4株瘤细胞的生长均有较明显的抑制作用 ,且呈剂量依赖关系。另外 ,在剂量相同的情况下 ,nrhTNF α的抑瘤作用明显强于重组人TNF α(rhTNF α)。结论nrhTNF α对小鼠移植性肿瘤生长具有很强的抑制作用。本研究为nrhTNF α临床的应用提供了重要的实验依据
Objective To observe the inhibitory effect of a novel genetically engineered human tumor necrosis factor α (nrhT NF α) on the growth of mouse transplanted tumors (sarcoma S180, liver cancer H2 2, melanoma B16 and Lewis lung cancer). Methods Mice were injected intramuscularly with nrhTNF α at 3 doses (37.5, 75 and 1.5 million U / kg body weight) once daily for 8 days. The inhibitory effect of nrhTNF α on the tumor cell line was observed by weighing the formed tumor weight. Results The calculated tumor inhibition rate showed that nrhTNF α had a significant inhibitory effect on the growth of 4 tumor cells in a dose-dependent manner. In addition, the anti-tumor effect of nrhTNFα was significantly stronger than recombinant human TNFα (rhTNFα) at the same dosage. Conclusion nrhTNF α has a strong inhibitory effect on the growth of transplanted tumors in mice. This study provides an important experimental basis for the clinical application of nrhTNFα
