目的 应用免疫组化法 (ABC法 )检测 ICA,比较研究 A型、B型、AB型与 O型 4种不同血型成人胰腺抗原底物对 ICA检测的特异性和敏感性的影响。方法 取 2 4例 1型糖尿病病人的 ICA阳性不同 JDF单位的血清标本和 2 3例正常人 ICA阴性血清标本 ,以 A型、B型、AB型与 O型 4种不同血型成人胰腺的冰冻切片为抗原底物 ,应用 ABC免疫组化法检测 ICA。结果 以 O型血成人胰腺为对照 ,应用 3例 A型和 B型血胰腺和 2例 AB型血胰腺为抗原底物进行ICA检测 ,2 4例 ICA阳性血清和 2 3例 ICA阴性血清检测结果的特异性和敏感性基本一致 ,未见假阳性或假阴性结果。结论 本文研究结果证明 A型、B型或 AB型血成人胰腺与 O型血成人胰腺一样 ,可用于 ICA免疫组化 (ABC法 )检测 ,不同血型人胰腺作为抗原底物不影响 ICA免疫组化法检测的特异性和敏感性。
Objective To detect ICA by immunohistochemistry (ABC method) and to compare the specificity and sensitivity of four different blood type human pancreatic antigen substrates (type A, type B, type AB and type O) to ICA detection. Methods Serum samples from 24 ICU positive patients with different types of diabetes mellitus (ICA) positive and two ICA negative serum samples from 23 normal individuals were collected and frozen sections of 4 adult human pancreas of type A, B, AB and O As antigenic substrate, ABC immunohistochemistry was used to detect ICA. Results O-type adult human pancreas was used as control, and 3 ICA-type and B-type blood pancreas and 2 type AB blood-pancreas were used as antigenic substrates to detect ICA, 24 ICA-positive sera and 23 ICA-negative sera The specificity and sensitivity are basically the same, no false positive or false negative results. Conclusion The results of this study demonstrate that type A, type B or type AB blood adult human pancreas can be used for ICA immunohistochemistry (ABC method) as well as for type 0 blood adult human pancreas. Different blood type human pancreas does not affect ICA immunohistochemistry Specificity and sensitivity of the assay.
