【目的】探讨肢体短暂缺血预处理对大鼠肺泡巨噬细胞(AMs)抑制作用。【方法】按常规建立大鼠肢体短暂远隔缺血预处理(RIPC)模型。实验设空白对照组(加入未行肢体缺血预处理大鼠血清1 mL),血清实验组(加入 RIPC 大鼠血清1 mL)和纳洛酮实验组(同时加入 RIPC 大鼠血清和10μg/mL 纳洛酮各0.5 mL)。酶联免疫法检测各组 AMs 吸光度 OD 值,计算 AMs 抑制率;流式细胞仪检测 AMs 凋亡情况。【结果】与空白对照组AMs 抑制率比较,血清实验组加入血清以后其抑制率达到42.5%( P <0.05),而纳洛酮实验组阻断了血清的这种效应[AMs 抑制率0%( P <0.05)]。流式细胞仪显示:血清实验组 AMs 凋亡率38.2%,明显高于空白组(0.92%)和纳洛酮组(0.91%)( P <0.05),而后两组组间差异无显著性( P >0.05)。【结论】RIPC 大鼠血清能抑制 AMs 生长、增殖,加速其凋亡,该抑制效应可被鸦片受体阻断剂纳洛酮阻断。“,”Objective]To investigate the inhibitory effect of limb transient ischemic preconditioning on alveo‐lar macrophage (AMs) in rats .[Methods]The modle of transient limb remote ischemia preconditions (RIPC)was routinely established .The experiment was set up in the blank control group (1 mL in the serum of the rats without limb ischemic preconditioning) ,the serum experimental group (1 mL rats were added to RIPC) and the naloxone experimental group ( 0 .5 mL serum of RIPC rats and 10 g/mL naloxone were added at the same time) .The OD value of AMs were detected by enzyme linked immunosorbent assay ,the AMs inhibition rate was calculated ;ap‐optosis of AMs was detected by flow cytometry .[Results]Compared with the blank control group ,the inhibition rate of AMs after adding serum was 42 .5% ( P < 0 .05) ,the naloxone group blocked the effect of serum (AMs in‐hibition rate was 0% ) ( P < 0 .05) .The flow cytometry showed that the apoptosis rate of serum AMs was 38 .2% , significantly higher than that in the blank group (0 .92% ) and the naloxone group (0 .91% ) ( P 0 .05) .[Conclusion] RIPC rat serum can inhibit the growth and proliferation of AMs and accelerate its apoptosis .The inhibitory effect could be blocked by naloxone , the opioid receptor antagonist .
