目的探讨端粒酶活性与宫颈癌及子宫颈上皮内瘤样变(CIN)的相关性。方法对50例浸润型宫颈癌,152例CIN(CINⅢ级58例、CINⅡ级44例、CIN级Ⅰ50例)和66例正常者的宫颈新鲜组织,用端粒酶重复扩增法-聚合酶链反应(TRAP-PCR)检测端粒酶活性。结果宫颈癌组中,端粒酶阳性42例(84.0%);CINⅢ级中端粒酶阳性33例(56.9%);CINⅡ级中,端粒酶阳性15例(34.1%);CINⅠ级中,端粒酶阳性12例(24.0%)。而66例正常宫颈组织仅有3例(4.55%)端粒酶活性表达,宫颈癌和CINⅢ组织端粒酶活性表达差异有统计学意义(P<0.01),CINⅢ级与正常对照组织端粒酶活性表达差异有统计学意义(P<0.01)。结论端粒酶活性在宫颈癌发生中可能起到重要作用,在子宫颈病变中检测端粒酶活性,对探讨宫颈病变的进展和宫颈癌的发生具有重要意义。
Objective To investigate the correlation between telomerase activity and cervical cancer and cervical intraepithelial neoplasia (CIN). Methods Fifty cases of invasive cervical cancer, 152 cases of CIN (58 cases of CIN Ⅲ grade, 44 cases of CIN Ⅱ grade, 50 cases of CIN grade Ⅰ) and 66 cases of normal fresh tissue of the cervix, using telomerase repeat amplification - polymerase chain Response (TRAP-PCR) to detect telomerase activity. Results The positive rate of telomerase was 42 (84.0%) in cervical cancer group, 33 (56.9%) were positive for telomerase in CINⅢ, 15 (34.1%) were positive in CINⅡ, Telomerase positive in 12 cases (24.0%). However, telomerase activity was only found in 66 cases of normal cervical tissue (4.55%), while there was a significant difference of telomerase activity between cervical cancer and CINⅢ (P <0.01). The expression of telomerase The difference of active expression was statistically significant (P <0.01). Conclusion Telomerase activity may play an important role in the development of cervical cancer. Detection of telomerase activity in cervical lesions is of great significance in exploring the progress of cervical lesions and cervical cancer.
