目的探讨骨髓间充质干细胞(bMSC)移植对大鼠UUO模型肾小管上皮细胞超微结构的改善作用及其可能的作用机制。方法体外分离培养雄性Wistar大鼠bMSC,体外Brdu标记,将Brdu-bMSC注入同种大鼠体内。45只受体大鼠随机分为3组:假手术对照组(SH组,n=15)、Brdu-bMSC+UUO组(BM组,n=15)、生理盐水+UUO组(NS组,n=15)。分别在3 d、7 d和14 d处死大鼠,取下肾脏组织,电镜观察肾小管上皮细胞微结构变化。采用免疫组织化学方法检测Brdu阳性bMSC。结果电镜下,随着结扎时间延长肾小管上皮细胞微结构破坏逐渐加重,术后3 d、7 d BMSC组肾小管上皮细胞超微结构明显改善;未在大鼠肾脏组织中发现Brdu阳性细胞。结论 bMSC移植可短期内改善肾小管上皮细胞超微结构,对肾小管间质病变起到保护作用,其作用机制可能是通过旁分泌作用。
Objective To investigate the effect of bone marrow mesenchymal stem cells (bMSC) transplantation on the ultrastructure of renal tubular epithelial cells in rat UUO model and its possible mechanism. Methods Male Wistar rats bMSCs were isolated and cultured in vitro. Brdu labeled in vitro and Brdu-bMSC were injected into the same kind of rats. 45 rats were randomly divided into 3 groups: sham operation control group (SH group, n = 15), Brdu-bMSC + UUO group (BM group, n = 15), saline + UUO group = 15). The rats were sacrificed on days 3, 7, and 14, respectively. The kidneys were removed and the changes of renal tubular epithelial cells were observed by electron microscope. Brdu-positive bMSCs were detected by immunohistochemistry. Results Under electron microscope, the destruction of renal tubular epithelial cells gradually increased with the extension of ligation time. The ultrastructures of renal tubular epithelial cells in BMSC group were significantly improved on the 3rd and 7th day after operation. Brdu positive cells were not found in the rat kidney. Conclusions Transplantation of bMSC can improve the ultrastructure of renal tubular epithelial cells in a short time and protect the tubulointerstitial lesion. Its mechanism may be through paracrine effect.
