目的探讨脑出血大鼠血肿周围组织中白细胞介素(IL)-17与核因子(NF)-κBp65的表达及其意义。方法 90只大鼠随机分成对照组,假手术组,出血组,每组30只。选取6、24 h,3、7、14 d五个时间点。RT-PCR法检测大鼠血肿周围组织中IL-17 m RNA,NF-κBp65 m RNA的表达。结果对照组与假手术组IL-17 m RNA与NF-κBp65 m RNA的表达在各时间点均无统计学差异(P>0.05)。在同一时间点,出血组IL-17 m RNA与NF-κBp65 m RNA的表达与对照组、假手术组均具有统计学差异(P<0.05)。在各时间点,出血组IL-17 m RNA、NF-κBp65 m RNA的表达均具有统计学差异(P<0.05);出血组IL-17 m RNA与NF-κBp65 m RNA的表达在脑出血后7 d高于其他时间点(P<0.05)。结论 IL-17 m RNA与NF-κBp65 m RNA的表达在脑出血后6 h就开始增加,7 d高于6、24 h,3、14 d。IL-17与NF-κBp65参与脑出血后的炎症反应。
Objective To investigate the expression and significance of interleukin (IL) -17 and NF-κBp65 in the perihematoma after intracerebral hemorrhage in rats. Methods Ninety rats were randomly divided into control group, sham operation group and hemorrhage group, 30 rats in each group. Select 6,24 h, 3,7,14 d five time points. The expression of IL-17mRNA and NF-κBp65mRNA in the perihematoma tissues of rats were detected by RT-PCR. Results The expression of IL-17 m RNA and NF-κBp65 m RNA in control group and sham operation group had no statistical difference at all time points (P> 0.05). At the same time point, the expression of IL-17mRNA and NF-κBp65mRNA in hemorrhage group were significantly different from those in control group and sham operation group (P <0.05). At each time point, the expressions of IL-17mRNA and NF-κBp65mRNA in the hemorrhage group were significantly different (P <0.05). The expressions of IL-17mRNA and NF-κBp65mRNA in the hemorrhage group were significantly higher than those in the hemorrhagic group 7 d higher than other time points (P <0.05). Conclusion The expression of IL-17mRNA and NF-κBp65mRNA began to increase 6 h after ICH, and was higher than 6,24 h and 3,14 d after 7 d. IL-17 and NF-κBp65 participate in the inflammatory reaction after intracerebral hemorrhage.
