目的:探讨ATP敏感钾通道(K_(ATP))与逼尿肌功能变化之间的关系。方法:根据膀胱压力容积测定结果将动物分为稳定组(29只)和不稳定组(9只),其中稳定组分为BOO(11只)、SCI模型(9只)和对照组(9只),通过RT-PCR技术测定逼尿肌中各型K_(ATP)mRNA表达及变化。结果:正常大鼠中逼尿肌不稳定发生率为24.3%。K_(ATP)在各组模型大鼠逼尿肌中均有表达。K_(ATP)的SUR2B亚型在BOO和SCI组逼尿肌中的表达高于正常对照组;IDI组中SUR2B的含量与正常对照组无明显差别。结论:K_(ATP)的表达异常可能是导致平滑肌细胞静息电位降低、兴奋性增高,引起逼尿肌不稳定的重要原因。
Objective: To investigate the relationship between ATP-sensitive potassium channel (K ATP) and detrusor function. Methods: The animals were divided into stable group (n = 29) and unstable group (n = 9) according to the measurement results of bladder pressure. The stable components were BOO (11), SCI model (9) and control group ). The expression and changes of K (ATP) mRNA in detrusor were determined by RT-PCR. Results: The incidence of detrusor instability in normal rats was 24.3%. K_ (ATP) was expressed in the detrusor of rats in each group. The expression of SUR2B subunit of K_ (ATP) in detrusor muscle of BOO and SCI group was higher than that of normal control group. The content of SUR2B in IDI group was not significantly different from that of normal control group. CONCLUSION: The abnormal expression of K ATP may be the important reason for the decrease of resting potentials and excitability of smooth muscle cells and the detrusor instability.
