本文对刀豆素A(ConA)在小鼠胰岛移植模型上的抗排异作用进行了研究。用胶原酶消化中手挑法获取C57BL/6小鼠的胰岛,将600—700个胰岛移植于链脲霉素复制成糖尿病的BALB/C小鼠肾包膜下,移植胰岛平均存活时间(MST)为14.1±2.5天。实验组用ConA对胰岛预处理后再移植,MST延长为25±5.4天,与对照比较有显著性(P<0.0005):给受鼠注射ConA也使MST显著延长(P<0.05),用ConA对胰岛及受鼠联合处理,MST延长为25.2±4.4天(P<0.0005);实验证明ConA有显著抑制排异反应的作用。对ConA作用机理的初步研究表明:荧光素标记的ConA能与胰岛良好结合,有可能覆盖组织相容抗原,注射ConA后,小鼠淋巴细胞对同系小鼠淋转反应显示一定的抑制作用。
In this paper, the anti-rejection effect of ConA on mouse islet transplantation model was studied. The islets of C57BL/6 mice were harvested by collagenase digestion and the 600-700 islets were transplanted under the renal capsule of BALB/C mice with the replication of streptozotocin. The average survival time of transplanted islets (MST) ) It was 14.1 ± 2.5 days. In the experimental group, ConA was used to pretreat the pancreatic islets before transplantation. The MST was prolonged to 25±5.4 days, which was significant compared with the control (P<0.0005). ConA was also given to the recipients and the MST was significantly prolonged (P<0.05). ConA was used. The combination of islets and rats, the MST prolonged to 25.2±4.4 days (P<0.0005). Experiments showed that ConA had significant inhibition of rejection. The preliminary study on the mechanism of ConA showed that Fluorescein-labeled ConA can bind well with islet, and it may cover histocompatibility antigen. After ConA injection, lymphocyte in mice showed certain inhibitory effect on lymphocyte reaction in syngeneic mice.
