目的 :观察通脑灵对缺氧条件下培养的神经细胞 bcl- 2和 bax基因表达的影响。方法 :采用原位杂交检测 bcl- 2 m RNA和 bax m RNA。结果 :通脑灵组 bcl- 2的表达在复氧后 6 h出现 ,2 4h及 48h阳性信号逐渐增强 ,而对照组在复氧 6 h出现 bcl- 2阳性信号 ,但随复氧时间的延长逐渐下降。通脑灵组和对照组 bax的表达均于复氧后6 h出现阳性信号 ,2 4h达高峰 ,48h逐渐下降。结论 :通脑灵的神经保护作用可能部分地与调控缺氧损伤神经细胞内 bcl- 2和 bax基因的表达有关
Objective : To observe the effect of Tongnaoling on the expression of bcl-2 and bax genes in cultured neurons under hypoxic conditions. Methods: In situ hybridization was used to detect bcl-2 mRNA and bax m RNA. RESULTS: The expression of bcl-2 in Tongnaoling group occurred 6 h after reoxygenation, and the positive signals at 24 h and 48 h gradually increased. However, bcl-2 positive signal appeared at 6 h after reoxygenation in control group, but the time of reoxygenation prolonged. decreasing gradually. The expression of bax in Tongnaoling group and control group showed positive signals 6 h after reoxygenation, peaked at 24 h and gradually decreased at 48 h. Conclusion : The neuroprotective effect of Tongnaoling may be partly related to the regulation of the expression of bcl-2 and bax genes in hypoxic injured nerve cells.
