                                目的:观察类风湿关节炎(RA)患者血浆中TNF-α、IL-1β及IL-10的表达情况,探讨TNF-α、IL-1β、IL-10与RA疾病活动度指标血浆ESR、CRP水平及DAS28评分的相关关系。方法:30例RA患者和10例健康志愿者,采集相关临床资料,检测C反应蛋白(CRP)及血沉(ESR)等,并进行DAS28评分。ELISA检测血浆中TNF-α、IL-1β及IL-10的表达,分析炎性细胞因子与疾病活动度指标的相关关系。结果:RA活动组患者血浆TNF-α、IL-1β水平明显高于健康对照组(P<0.05),IL-10水平有显著下降(P<0.05)。RA缓解组TNF-α、IL-10水平较健康对照组有明显升高,(P<0.05),IL-1β无明显变化(P>0.05)。相关性分析结果显示TNF-α、IL-1β与ESR、CRP水平及DAS28评分呈正相关(P<0.05),IL-10与ESR、CRP水平及DAS28评分无明显相关。结论:RA患者血浆中TNF-α、IL-1β及IL-10可作为疾病活动度的重要参考指标。
Objective: To observe the expression of TNF-α, IL-1β and IL-10 in the plasma of patients with rheumatoid arthritis (RA), and to explore the plasma ESR, CRP Correlation between levels and DAS28 score. Methods: Thirty patients with RA and 10 healthy volunteers were enrolled in this study. Relevant clinical data were collected and C-reactive protein (CRP) and erythrocyte sedimentation rate (ESR) were measured. The DAS28 score was also used. The levels of TNF-α, IL-1β and IL-10 in plasma were detected by ELISA. The correlation between inflammatory cytokines and disease activity index was analyzed. Results: The levels of plasma TNF-α and IL-1β in patients with active RA were significantly higher than those in healthy controls (P <0.05) and IL-10 levels were significantly decreased (P <0.05). The level of TNF-α and IL-10 in RA remission group was significantly higher than that in healthy control group (P <0.05), while there was no significant change in IL-1β (P> 0.05). Correlation analysis showed that there was a positive correlation between TNF-α, IL-1β and ESR and CRP levels and DAS28 score (P <0.05). There was no significant correlation between IL-10 and ESR, CRP level and DAS28 score. Conclusion: The levels of TNF-α, IL-1β and IL-10 in plasma of RA patients may be used as important reference indexes for disease activity.
