目的:探讨清肺培元颗粒对免疫低下肺部感染小鼠的可能作用机制。方法:建立免疫低下肺部感染小鼠模型,随机分为8组,各组给药14d后摘取眼球取静脉血;流式细胞术检测小鼠外周血中CD_4~+T、CD_8~+T细胞含量,计数CD_4~+/CD_8~+的比值;ELISA法测定小鼠血清中白细胞介素6(IL-6)及降钙素(PCT)含量。结果:各治疗组病理学改变较模型组轻,中西药组,清肺培元颗粒中、高剂量组改善明显;与正常对照组比较,模型组CD4+显著降低(P<0.05),CD_8~+显著升高(P<0.05);与模型组比较,用药后清肺培元颗粒3个剂量组小鼠,CD_4~+T淋巴细胞水平及CD_4~+/CD_8~+比值明显升高,同时降低CD_8~+T淋巴细胞数量(P<0.05);模型小鼠血清中IL-6、PCT表达水平均较正常组增高(P<0.05),各治疗组较模型组显著降低(P<0.01)。结论:清肺培元颗粒有效地调节小鼠外周血T淋巴细胞亚群的水平,降低IL-6、PCT的表达水平,能够增强机体免疫功能。
Objective: To investigate the possible mechanism of Qingfei Peiyuan Granule on mice with immunocompromised pulmonary infection. Methods: Mice with immunocompromised pulmonary infection were established and randomly divided into 8 groups. The rabbits in each group were sacrificed for 14 days and their venules were removed. The levels of CD_4 ~ + T and CD_8 ~ + T in peripheral blood were detected by flow cytometry The ratio of CD_4 ~ + / CD_8 ~ + was counted and the levels of IL-6 and PCT in serum were measured by ELISA. Results: The pathological changes in each treatment group were lighter than those in the model group. The levels of CD4 + in the model group were significantly lower than those in the normal control group (P <0.05) (P <0.05). Compared with the model group, the levels of CD_4 ~ + T lymphocytes and the ratio of CD_4 ~ + / CD_8 ~ + of mice in the three doses of Qingfei Peiyuan Granules were significantly increased (P <0.05). The levels of IL-6 and PCT in the model mice were significantly higher than those in the normal group (P <0.05). The levels of CD_8 ~ + T lymphocytes were significantly decreased in all treatment groups compared with the model group (P <0.01). Conclusion: Qingfei Peiyuan Granule can effectively regulate the level of T lymphocyte subsets in peripheral blood and reduce the expression of IL-6 and PCT, which can enhance the immune function.
