目的 :观察生长抑素对人胰腺癌细胞株HS76 6T生长的作用 ;测定神经鞘磷脂 (sphingomyolin ,SM )、cAMP、细胞内Ca2 + 水平 ,探讨生长抑素的受体后信息传递作用。方法 :细胞培养 ,分别加入不同浓度的生长抑素 ,PACAP1-3 8。应用MTT法来观察细胞增殖程度 ;薄层层析法测定细胞内SM ,放免法测定细胞内cAMP含量 ;Fura - 2 /AM测定 [Ca2 + ]i。结果 :不同浓度的生长抑素 (10 -6~ 10 -12 mol/L)均能有效地抑制HS76 6T的生长。能抑制HS76 6T细胞内SM、cAMP生成和细胞内Ca2 + 水平 ,cAMP生成量和细胞内Ca2 + 水平与生长抑素浓度呈负相关。结论 :生长抑素能抑制HS76 6T细胞的生长 ,它的抑制作用是通过受体介导的
OBJECTIVE: To observe the effect of somatostatin on the growth of human pancreatic cancer cell line HS76 6T, to determine the level of sphingomyolin (SM), cAMP and intracellular Ca2 +, and to explore the effect of somatostatin on post-receptor information transmission. Methods: Cell culture was added different concentrations of somatostatin, PACAP1-3 8. The degree of cell proliferation was observed by MTT assay. The content of intracellular cAMP was determined by TLC and the [Ca2 +] i was determined by Fura - 2 / AM. Results: Different concentrations of somatostatin (10 -6 ~ 10 -12 mol / L) could effectively inhibit the growth of HS76 6T. Could inhibit the production of SM and cAMP and the level of intracellular Ca2 + in HS76 6T cells. The production of cAMP and the level of intracellular Ca2 + were negatively correlated with the concentration of somatostatin. Conclusion: Somatostatin can inhibit the growth of HS76 6T cells and its inhibitory effect is mediated by receptor
