目的β-细辛醚对核转录因子κB(nuclear factor Kappa B,NF-κB)表达的影响,为进一步应用开发打下基础。方法培养RAW264.7细胞,脂质体LipofectamineTM2000转染NF-κB启动子双荧光素酶报告基因分析系统Luc2P/NF-κB-RE/Hygro,0.2μg/mL LPS刺激,以15、150μmolβ-细辛醚干预,孵育24 h后检测相对荧光素酶活性;按以上分组干预培养RAW264.7细胞,采用蛋白免疫印迹法检测NF-κB蛋白表达。结果荧光素酶报告基因结果表明,β-细辛醚有抑制NF-κB启动子活性作用,且随剂量增大抑制作用增强;蛋白免疫印迹法结果表明,β-细辛醚有抑制NF-κB蛋白表达作用,且随剂量增大抑制作用增强。结论β-细辛醚能抑制NF-κB转录与表达,可能有较强的抗炎作用。
Objective To study the effect of β-asarone on the expression of nuclear factor kappa B (NF-κB) in order to lay a foundation for further application and development. METHODS: RAW264.7 cells were cultured and transfected with Luciferase reporter assay Luciferase reporter assay (Luc2P / NF-κB-RE / Hygro, 0.2μg / mL LPS) Ether for 24 h, then the relative luciferase activity was measured. The RAW264.7 cells were cultured in the above groups, and the protein expression of NF-κB was detected by Western blotting. Results The results of luciferase reporter assay showed that β-asarone could inhibit the activity of NF-κB promoter and its inhibitory effect increased with increasing dose. Western blotting showed that β-asarone could inhibit NF-κB Protein expression, and with increasing dose inhibition increased. Conclusion β-asarone can inhibit the transcription and expression of NF-κB and may have a strong anti-inflammatory effect.
