目的研究河北省2005-2008年甲3亚型流感病毒HA1基因变异特征。方法选取河北省2005-2008年分离的甲3亚型流感病毒进行核酸提取,采用逆转录-聚合酶链反应(RT-PCR)扩增病毒HA1基因后进行核苷酸序列测定,用生物信息软件对测序结果进行分析处理。结果2005-2006年分离株与当年疫苗株A/California/7/2004比较,在其HA1蛋白分子上有6个氨基酸位点发生替换;2006-2007年分离株与当年疫苗株A/Hiroshima/52/2005比较,有4个氨基酸位点发生替换;2007-2008年分离株与当年疫苗株A/Wisconsin/67/2005比较,有8个氨基酸位点发生替换,且与前两个流行期的分离株比较存在4、6个氨基酸位点差异。基因进化树上,同一流行期分离株基本呈集中分布,每个流行期分离株均远离当年疫苗株。结论河北省2005-2008年分离的甲3亚型流感病毒HA1基因发生了变异,在HA1氨基酸序列的进化亲缘关系结构上具有同流行期相近的特征。
Objective To study the variation of HA1 gene of influenza virus type 3 in Hebei Province from 2005 to 2008. Methods Nucleic acid was extracted from Influenza virus subtype A (H3 subtype) isolated in Hebei Province during 2005-2008. The nucleotide sequence of HA1 gene was amplified by reverse transcription-polymerase chain reaction (RT-PCR). The bioinformatics software The sequencing results were analyzed and processed. Results There were 6 amino acid substitutions in the HA1 protein between 2005-2006 isolates and the vaccine strain A / California / 7/2004. The isolates from 2006-2007 and the vaccine strain A / Hiroshima / 52 / 2005 compared to 4 amino acid sites were replaced; 2007-2008 isolates and vaccine year strain A / Wisconsin / 67/2005 compared to 8 amino acid sites were replaced, and the first two popular period of separation There are 4 and 6 amino acid differences in the strains. On the phylogenetic tree, the isolates in the same epidemic period were basically concentrated, and the isolates in each epidemic period were far away from the vaccine strains of the year. Conclusion HA1 gene of influenza A virus isolated from 2005 to 2008 in Hebei Province has been mutated. The phylogenetic relationship of amino acid sequence of HA1 is similar to that of HA1 gene.
