目的 观察修饰有骨形态发生蛋白质7(bone morphogenetic protein 7,BMP-7)功能片段的功能化自组装多肽纳米纤维水凝胶RADKPS在低浓度炎症因子白介素-1β(inter leukin-1β,IL-1β)(10 pg/ml)持续作用下对大鼠髓核细胞蛋白多糖和II型胶原代谢的影响.方法 体外分离培养SD大鼠髓核细胞(nucleus pulposus cells,NPCs),取第3代NPCs分别在无BMP-7功能片段的RADA16-I或修饰有BMP-7功能片段的RADKPS中培养,试验共分4组:A组(无IL-1β 干预的RADA16-I组)、B组(有IL-1β 干预的RADA16-I组)、C组(有IL-1β 干预的RADKPS组)和D组(无IL-1β 干预的RADKPS组).分别在培养的第3天、第6天及第9天通过RT-PCR和ELISA检测髓核细胞蛋白多糖和II型胶原在RNA和蛋白水平的表达量.结果 RT-PCR和ELISA结果表明在第3天和第6天时C组蛋白多糖和II型胶原的表达水平均明显高于A组,差异均有统计学意义(P0.05);B组蛋白多糖和II型胶原的表达水平在第3天、第6天及第9天时均低于A组,但差异均无统计学意义(P>0.05);在第3天时C组蛋白多糖和II型胶原的表达水平低于D组,差异无统计学意义(P>0.05),在第6天和第9天时,C组中两者的表达量均明显低于D组,且差异均有统计学意义(P 0.05 ); the expression level of aggrecan and type Ⅱ collagen of group C was lower than that of group D on the 3rd day and there was no statistical signiifcance (P> 0.05 ). But the expression level of aggrecan and type Ⅱ collagen of group C was signiifcantly lower than that of group D on the 6th day and the 9th day, and the difference between the 2 groups was statistically signiifcant (P < 0.05 ).Conclusions After being stimulated by BMP-7 functional fragments, nucleus pulposus cells are more sensitive to the inhibition of IL-1β on the synthesis of aggrecan and type Ⅱ collagen. However, IL-1β ( 10 pg / ml ) could not completely counteract the positive effect of BMP-7 functional fragments which are compounded in the functionalized self-assembled peptide nanoifber hydrogel RADKPS on the synthesis of aggrecan and type Ⅱ collagen of NPCs. The present study shows that RADKPS has a certain anti-IL-1β effect in vitro. Therefore, RADKPS can still promote the synthesis of aggrecan and type Ⅱ collagen of NPCs when proinflammatory cytokine IL-1β is present.
