目的:研究苦参碱对视网膜母细胞瘤(Rb)细胞增殖与凋亡及PI3K/Akt信号通路的影响.方法:将Rb细胞株Y79作传代培养,使用不同浓度(15、30、60、120mg/L)的苦参碱对细胞进行培养作为观察组,同时选择空白组作为对照组,采用噻唑蓝比色法(MTT)检测生长抑制情况,流式细胞术检测细胞凋亡情况,计算凋亡率,RT-PCR法检测PI3K、AktmRNA相对表达量,免疫组化法检测PI3K、Akt蛋白表达情况.结果:培养24小时、48小时、72小时后,观察组各组Y79细胞生长抑制率均高于对照组,并且随着时间与苦参碱浓度的增加,生长抑制率也在升高(P<0.05);经过培养,观察组在G0/G1期的细胞比例高于对照组,处于S期和G2/M期细胞低于对照组(P<0.05);相同时间内,随着苦参碱浓度的升高,细胞凋亡率明显升高,且高于对照组(P<0.05).相同浓度时,随着时间的增加,细胞凋亡率也升高,均高于同时期对照组(P<0.05);观察组中各浓度条件下,PI3KmRNA、AktmRNA相对表达量均明显低于对照组(P<0.05),并且随着浓度的升高,PI3KmRNA、AktmRNA相对表达量显著降低(P<0.05);观察组中各浓度条件下,PI3K、Akt蛋白表达均低于对照组(P<0.05),并且随着浓度的升高,PI3K、Akt蛋白表达也下降(P<0.05).结论:苦参碱能够明显抑制Rb细胞的增殖,促进细胞凋亡,可能是通过抑制PI3K/Akt信号通路,使细胞停留在G0/G1期,诱导细胞凋亡.“,”Objective:To investigate the effect of Matrine on proliferation, apoptosis and PI3 K/Akt signaling pathway for retinoblastoma cells.Methods:The Rb cell line Y79 were cultured, using different concentrations (15, 30, 60, 120 mg/L) of matrine were cultured as the observation group of cells, and the blank group as the controlled group, MTT method (MTT) to detect growth inhibition, cell apoptosis by flow cytometry.The apoptosis rate was calculated by RT-PCR detection of PI3 K, the relative expression of AktmRNA and PI3 K was detected by immunohistochemical method, the expression of Akt protein.Results:After culturing 24 h, 48 h and 72 h, the growth inhibition rate of Y79 cells in the observation group was higher, and the growth inhibition rate was also increased with the increase of time and matrine concentration (P < 0.05).After training, the proportion of cells in the observation group in GO/G1 phase was higher, and the cells in phase S and G2/M phase were lower (P < 0.05).At the same time, with the increase of matrine concentration, the apoptosis rate increased significantly and higher (P < 0.05).The relative expressions of PI3 KmRNA and AktmRNA in the observation group were significantly lower, and with the increase of concentration, the relative expression of PI3 KmRNA and AktmRNA decreased significantly (P < 0.05).In the observation group, the expressions of PI3 K and Akt proteins were lower, and with the increase of concentration, the expression of PI3 K and Akt protein also declined (P < 0.05).Conclusion:Matrine can significantly inhibit the proliferation of Rb cells and promote apoptosis, which may cause the cells to stay in the GO/G1 phase and induce apoptosis by inhibiting the PI3 K/Akt signaling pathway.
