OBJECTIVE: To determine the expression level of uncoupling protein-2 (UCP2) mRNA in rat skeletal muscle by high-calcium intervention in early-weaned rats and high fat diet in adult rats, and to investigate the effect of high calcium intake on early obesity The formation of the impact. Methods 120 severed male Wistar rats were randomly divided into normal diet group and high calcium diet (2%), middle (2.5%) and high dose (3%) groups. After 4 weeks of high calcium intervention, blood lipids were measured. All rats were subsequently fed a basal diet for 3 weeks. At the end of the seventh weekend blood test for lipid. The rats of normal diet were randomly divided into normal control group and obesity induction group. Fat-induced group and high calcium group were fed with high-fat diet. After 8 weeks of obesity induction, all rats were sacrificed to measure blood lipids and cryopreserved Skeletal muscle RT-PCR semi-quantitative detection of UCP 2 gene expression levels. Results After high-fat diet, the body weight gain of three high-calcium groups was lower than that of the obesity-induced group. There was no difference between the high-fat group and the normal control group. The high-dose group had significantly lower serum triglyceride Obesity-induced group, consistent with the normal control group; obesity-induced group and high-dose group UCP 2 gene expression was significantly lower than the normal control group, low dose and middle dose group; medium dose of high calcium group expression was significantly higher than the normal control group. Conclusion The high calcium diet in weaned rats can continue to play a role in enhancing the expression of UCP2 gene and improving dyslipidemia during the adult high fat diet.
