目的 探讨钙离子 (Ca2 + )和温度对肿瘤细胞粘附能力的影响。 方法 利用细胞培养技术 ,以K ras基因转染的鼠肺纤维 (KNRL)细胞———一种恶性转化细胞做为研究对象 ,以其来源的鼠肺纤维 (NRL)细胞做为对照 ,在细胞培养液内加入Ca2 + 及分别在 4、37及 41℃状况下进行培养 ,观察对比 2种细胞聚集能力的改变。 结果 37℃细胞培养液内无Ca2 + 时 ,KNRL细胞的聚集能力(Nt/N0 )的均值为 0 81± 0 15 ,培养液内加入Ca2 + 时均值为 0 6 0± 0 14,两者差异有显著意义 (t =2 32 ,P <0 0 5 ) ;41℃下培养 ,KNRL细胞的聚集能力 (Nt/N0 )均值为 0 5 3± 0 12与 37℃下培养相比 ,差异有显著意义 (t=2 13 ,P <0 0 5 )。 结论 适当增加肿瘤细胞间Ca2 + 浓度和适当的高温 ,有助于提高肿瘤细胞的粘附能力
Objective To investigate the effect of Ca2 + and temperature on the adhesion of tumor cells. METHODS: The murine lung fiber (KNRL) cells, a type of malignant transformed cell line transfected with K ras gene, were used as the research object. The murine lung fiber (NRL) cells were used as a control in cell culture. Ca2 + was added into the culture medium and cultured under the conditions of 4, 37 and 41 ℃, respectively. The changes of the aggregation ability of the two kinds of cells were observed and compared. Results The average Nt / N0 concentration of KNRL cells was 0 81 ± 0 15 at 37 ℃ without Ca2 + and the average value of Ca2 + was 0 6 0 ± 0 14 (T = 2 32, P <0 05). Compared with the culture at 37 ℃, the average ability of KNRL cells to aggregate (Nt / N0) at 41 ℃ was significantly higher than that at 37 ℃ Significance (t = 2 13, P <0 0 5). Conclusions Appropriate increase of intercellular Ca2 + concentration and appropriate high temperature may help to improve the adhesion of tumor cells
