目的 观察葡萄糖转运体(GLUT1、GLUT3)在脑胶质瘤中的表达及其意义。方法应用 Northern blot技术检测不同级别脑胶质瘤葡萄糖转运体 mRNA表达情况,用 Western blot检测葡萄糖转运体的蛋白水平。结果 Ⅰ/Ⅱ级、Ⅲ级、Ⅳ级胶质瘤GLUT1 mRNA/肌动蛋白(actin)mRNA分别为0.12、0.17、0.21;4.2 kb GLUT3 mRNA/actin mRNA分别为0.178.0568、0.710;2.7kb GLUT3 mRNA/actin mRNA分别为0.075、0.216、0406。随着脑胶质瘤分级的增高,GLUT1/actin 转录物比率有随之增加的趋势,但无显著相关。4.2 kb GLUT3是2种转录物中丰度较高的一种,2种转录物的 GLUT3/actin比率与胶质瘤的分级呈线性正相关。结论 恶性胶质瘤细胞可能分泌某种因子增强 GLUTs mRNA表达,但抑制 GLUT1 mRNA翻译而促进 GLUT3蛋白表达,使GLUT3成为供应恶性胶质细胞能量代谢底物的主要载体。
Objective To observe the expression of glucose transporter (GLUT1, GLUT3) in glioma and its significance. Methods Northern blot was used to detect the mRNA expression of glucose transporter in glioma of different grades. The protein level of glucose transporter was detected by Western blot. Results The GLUT1 mRNA and actin mRNA of grade Ⅰ, Ⅱ, Ⅲ and Ⅳ were 0.12, 0.17 and 0.21, respectively. The expression of 4.2 kb GLUT3 mRNA / actin mRNA was 0.178.0568,0.710 and 2.7 kb GLUT3 mRNA / actin mRNA were 0.075,0.216,0406. With the increase of glioma grading, the ratio of GLUT1 / actin transcripts has an increasing trend, but no significant correlation. 4.2 kb GLUT3 is a more abundant one of the two transcripts. The GLUT3 / actin ratio of the two transcripts was positively correlated with the grade of glioma. Conclusions Glioblastoma cells may secrete some factor to enhance the expression of GLUTs mRNA, but inhibit the translation of GLUT1 mRNA to promote the expression of GLUT3 protein, making GLUT3 the main carrier for the energy metabolism of glioblastoma.
