目的:探讨欢乐宁方对围绝经期综合征小鼠下丘脑KNDy编码基因及其相关受体信号通路的影响。方法:建立11月龄ICR小鼠围绝经期综合征模型,随机分为模型组、欢乐宁方低、中、高剂量组、补佳乐组,另取2月龄小鼠作为正常组,每组各10只;测定各组小鼠血清E_2、LH、FSH水平,检测下丘脑组织Ki SS-1、NKB、DYN及相关受体GPR54、NK3和KOR mRNA的表达。结果:围绝经期小鼠血清E2明显降低,血清FSH、LH明显升高;Kiss-1、GPR54、NKB、NK3、DYN、KOR mRNA表达量明显上升。欢乐宁方能明显升高围绝经期小鼠血清E2水平,降低血清FSH、LH水平,能下降下丘脑Kiss-1、GPR54、NKB、NK3、DYN、KOR mRNA的表达,以中高剂量为佳。结论:欢乐宁方能有效调节围绝经期综合征小鼠的性激素水平;欢乐宁方可能通过作用于KNDy神经元,从而影响GnRH的分泌来调节H-P-O轴,这可能是欢乐宁方治疗围绝经期综合征的作用机制之一。
Objective: To investigate the effects of Huanleining-ning Fang on KNDy gene and its receptor pathway in hypothalamus of mice with peri-menopausal syndrome. Methods: A model of climacteric syndrome of 11-month-old ICR mice was established and randomly divided into model group, Huanleining low, medium and high dose groups, nociceptive group and 2-month-old mice as normal group. The serum levels of E_2, LH and FSH in each group were measured. The expressions of Ki SS-1, NKB, DYN and related receptors GPR54, NK3 and KOR mRNA in hypothalamus were detected. Results: Serum E2 of perimenopausal mice decreased significantly, serum FSH and LH were significantly increased; expression of Kiss-1, GPR54, NKB, NK3, DYN and KOR mRNA increased significantly. Huanleinning could significantly increase serum E2 level, decrease serum FSH and LH levels in perimenopausal mice, and decrease the expression of Kiss-1, GPR54, NKB, NK3, DYN and KOR in the hypothalamus. Conclusion: Huanleining could effectively regulate the sex hormone level in mice with perimenopausal syndrome. Huanleining could regulate the HPO axis by affecting the secretion of GnRH in KNDy neurons. This may be because Huanleining decoction can treat perimenopause One of the mechanisms of the syndrome.
